Getting ready to do my first 5K service this weekend and noticed in the owner's manual that it says the TPWS system must be reinitialized after every tire rotation, change etc. Does this actually trigger a relearn of tire position or just resets the baseline tire pressure readings?

I figured being a 2025 it would auto relearn positions after a few miles, but according to the way the manual reads it doesn't.

I have never thought about it or worried about it
It just syncs and works

I am at 25k miles and rotate tires between 3-5k

I just had a similar issue. I bought a set of the 18" TRD PRO wheels and Bridgestone Dueler tires for my TRD OR. I also got new TPMS sensors from Toyota and the black lugs to match the wheels. When I drove away from my tire shop the TPMS on the dash display was working fine, then about 50 miles later I had a warning going off that there was a "malfunction" in the system and I needed to contact my dealer.

I was due for my 10,000 mile service so instead of taking it back to the tire shop to complain I'd just see what the dealer said. It turns out the the truck stores the "signature" of the TPMS sensors that are installed in the wheels. I was told that buying new sensors meant that I had to have the system in the car recalibrated to recognize the new ones...half an hour and $170 later it was done. The tech at the dealer did tell me that now BOTH my wheel sets were stored in the car and if I swapped the original wheels back on the truck would automatically recognize them. He says Toyota sets it up like that for owners that swap wheel sets on and off based on what they need. I did not ask what happens with aftermarket sensors since I have the OEM ones.

?, sounds like you got ripped off by the dealership. I bought brand new wheels and tires, along with new TOYOTA, TPMS and yes the truck didn't recognize them until I went into the DASHBOARD settings, scroll to the TPMS setting and held down the button until it relearned the new sensors.
I will say I didn't understand what the manual was telling me to do, I only had the truck less than a month and really hadn't read the manual 😅. My solution was kinda the same, I went to the dealership and had someone in the service department show me how to do it for free.
